What can the MCU do now with Spider-Man ?

In Forest Hills, Queens, New York, high school student Peter Parker is a science-whiz orphan living with his Uncle Ben and Aunt May. He is bitten by a radioactive spider (erroneously classified as an insect in the panel) at a science exhibit and "acquires the agility and proportionate strength of an arachnid." Along with super strength, Parker gains the ability to adhere to walls and ceilings. Through his native knack for science, he develops a gadget that lets him fire adhesive webbing of his own design through small, wrist-mounted barrels. Initially seeking to capitalize on his new abilities, Parker dons a costume and, as "Spider-Man", becomes a novelty television star. However, "He blithely ignores the chance to stop a fleeing thief, [and] his indifference ironically catches up with him when the same criminal later robs and kills his Uncle Ben." Spider-Man tracks and subdues the killer and learns, in the story's next-to-last caption, "With great power there must also come—great responsibility!" Despite his superpowers, Parker struggles to help his widowed aunt pay rent, is taunted by his peers, particularly football star Flash Thompson, and engenders the editorial wrath of newspaper publisher J. Jonah Jameson. As he battles his enemies for the first time, Parker finds juggling his personal life and costumed adventures difficult. This is Marvel's chance to do Spider-Man right compared to what Sony has done to the character in their past movies.

In summary, an explosion in Stamford by the villain Nitro causes the U.S. Government to introduce the Superhero Registration Act. Those not adhering to it are deemed unregistered and rogue superheroes. Tony Stark and Reed Richards lead the side of the pro-registration superhero. However, Captain America leads the anti-registration side. Spider-Man, initially sided with Tony Stark, but eventually joins the team of Captain America after Goliath is killed by the pro-registration superheroes and their Thor Cyborg. Many of the super-villains join the Government in hunting down unregistered superheroes. Spider-Man is needed in this huge story line when Marvel Studios plans to tell this amazing tale.

Norman Osborn (reformed) became a public hero for his role in repelling the Skrull threat. Osborn, also given leadership of the espionage agency S.H.I.E.L.D., reforms that agency into H.A.M.M.E.R. and creates a new Avengers team under its aegis. The initial line-up consisted of former Thunderbolts members and new recruits, including the Sentry, Ares, Captain Marvel as well as disguised super-villains Moonstone (portraying as Ms. Marvel), Venom (Mac Gargan, portraying as Spider-Man after given a formula resetting the symbiote to the size when it possessed Spider-Man), Bullseye (portraying as Hawkeye) and Wolverine's disgruntled son Daken taking on his father's mantle. Osborn takes the identity of Iron Patriot, wearing an American themed Iron Man armor.

Norman Osborn is the original and most well-known incarnation of the Green Goblin. As an amoral industrialist at the head of Oscorp, he took a formula which enhanced his physical abilities and intellect, but also drove him to insanity. He adopted a Halloween-themed appearance, dressing in a goblin costume, riding on a bat-shaped "Goblin Glider", and using an arsenal of high-tech weapons, notably grenade-like "Pumpkin Bombs", to terrorize New York City. He is one of Spider-Man's most persistent archenemies that is responsible for numerous tragedies like Gwen Stacy's death, and is Harry Osborn's father. However, he has also come into conflict with other superheroes in the Marvel Universe, and was the focus of the company-wide Dark Reign storyline as the original Iron Patriot. For many years we had Harry Osborn mutated into a goblin, a ski-masked Harry Osborn, and Norman Osborn, who actually looked like an actual goblin, wearing a stupid motorcycle helmet. This is Marvel's final chance to have an actual Green Goblin in the movies.

Venom is a symbiote, a sentient alien, with a gooey, almost liquid-like form, and it requires a host, usually human, to bond with for its survival. After bonding, the symbiote endows its enhanced powers upon the host. When the Venom Symbiote bonds with a human, that new dual-life form refers to itself as "Venom." The Venom Symbiote's first known host was Spider-Man, who eventually separated himself from the creature when he discovered its true nature. The Symbiote went on to merge with other hosts, most notably Eddie Brock, its second and most famous host, with whom it first became Venom and one of Spider-Man's archenemies. According to S.H.I.E.L.D., it is considered to be one of the greatest threats to humanity, alongside Magneto, Doctor Doom, and Red Skull. When Sony introduced this character into Spider-Man 3, the character was completely ruined. Marvel now has a chance to make this character right, most likely in Marvel's Spider-Man 3.

After suffering three defeats in a row from Spider-Man, Doctor Octopus contacts every known super-villain that had ever crossed paths with Spider-Man. Only The Vulture, Electro, Kraven the Hunter, Mysterio, and Sandman had responded to his request. As none of the members are willing to relinquish the honor of killing Spider-Man themselves, they decide to challenge Spider-Man individually, with the order in which they face him determined by random drawing. The Sinister Six had kidnaped The Daily Bugle's secretary, Betty Brant, and Aunt May, holding them hostage in order to be able to force Spider-Man to confront them. Spider-Man defeats the Sinister Six one-by-one, mocking their decision to battle him individually instead of as a team. Sony had plans to be able to do this storyline. However, Marvel Studios will likely do this storyline possibly after Marvel's Spider-Man 3.
