Humans who had no understanding of biology or science saw significant differences between two large groups of humans and labeled them male and female, this ignored and penalized people who did not fall neatly into one or the other. Then, through religion, tradition, and violence, people forced behaviors, aesthetics, and personality traits onto these groups. Any person who was not interested in adhering to these behaviors, aesthetics, and personality traits was murdered, beaten, tortured, and/or raped. Prescriptive Gender Roles are violence which further punishes people for not falling neatly into imaginary boxes.

The understanding of chromosomal differences came after the assignment of male or female. The vast majority of people never have their chromosomes checked and assume their own gender. The only time this becomes a conflict is when someones gender doesn't fit the majorities expectations of gender based on arbitrary queues that are assigned based on prescriptive gender roles. People who lash out at transgender people don't behave in this manner because they really care about chromosomes, they do it because they are upset that their understanding of gendered queues is inadequate. They punish people because they are afraid that they are wrong about everything.
